Is orforglipron FDA-approved?
FDA-approved as Foundayo on April 1, 2026 for chronic weight management in adults. Taken once a day, with or without food: the label sets no wait before eating.
Overview
Orforglipron switches on the same GLP-1 receptor as the injectable medicines but is a small chemical that survives digestion. The FDA approved it as Foundayo in April 2026 for adults with obesity, or with overweight and at least one weight-related health problem, alongside diet and exercise. In the 72-week ATTAIN-1 trial in adults with obesity, the highest dose led to about 11% average weight loss, versus about 2% on placebo. The ACHIEVE trials test it in type 2 diabetes.
How it works
It binds the GLP-1 receptor and triggers the same effects as GLP-1 shots: more insulin when sugar is high, slower stomach emptying and less appetite.
How long does orforglipron stay in the body?
About 29 to 49 hours, so it builds to a steady level after about a week of daily pills. Levels peak 4 to 8 hours after a dose. About 77% of a dose is absorbed, and food lowers that only a little.
What it's studied for
- Obesity
- Type 2 diabetes
- Keeping weight off after stopping injectables
Reported side effects
- Nausea, constipation, diarrhea, vomiting, indigestion
- Slows the stomach, which can change how other pills are absorbed
- Boxed warning: thyroid C-cell tumors are a GLP-1 class warning. Orforglipron didn't cause them in rodents, but the label still rules it out for people with a personal or family history of medullary thyroid cancer or MEN 2.
How should orforglipron be stored?
Room temperature, in the original bottle.
Related: Oral semaglutide, Tirzepatide, Semaglutide, Mazdutide.
